Detecting and treating Alzheimer’s disease, before cognitive deficits occur, has become the health challenge of our time. The earliest known event in Alzheimer’s disease is rising amyloid-b. Previous studies have suggested that effects on synaptic transmission may precede plaque deposition. Here we report how relative levels of different soluble amyloid-b peptides in hippocampus, preceding plaque deposition, relate to synaptic and genomic changes. Immunoprecipitation-mass spectrometry was used to measure the early rise of different amyloid-b peptides in a mouse model of increasing amyloid-b (‘TASTPM’, transgenic for familial Alzheimer’s disease genes APP/PSEN1). In the third postnatal week, several amyloid-b peptides were above the limit of detection, including amyloid-b40, amyloid-b38 and amyloid-b42 with an intensity ratio of 6:3:2, respectively. By 2 months amyloid-b levels had only increased by 50% and although the ratio of the different peptides remained constant, the first changes in synaptic currents, compared to wild-type mice could be detected with patch-clamp recordings. Between 2 and 4 months old, levels of amyloid-b40 rose by 7-fold, but amyloid-b42 rose by 25-fold, increasing the amyloid-b42:amyloid-b40 ratio to 1:1. Only at 4 months did plaque deposition become detectable and only in some mice; however, synaptic changes were evident in all hippocampal fields. These changes included increased glutamate release probability (P50.001, n = 7–9; consistent with the proposed physiological effect of amyloid-b) and loss of spontaneous action potential-mediated activity in the cornu ammonis 1 (CA1) and dentate gyrus regions of the hippocampus (P5 0.001, n = 7). Hence synaptic changes occur when the amyloid-b levels and amyloid-b42:amyloid-b40 ratio are still low compared to those necessary for plaque deposition. Genome-wide microarray analysis revealed changes in gene expression at 2–4 months including synaptic genes being strongly affected but often showing significant changes only by 4 months. We thus demonstrate that, in a mouse model of rising amyloid-b, the initial deposition of plaques does not occur until several months after the first amyloid-b becomes detectable but coincides with a rapid acceleration in the rise of amyloid-b levels and the amyloid-b42:amyloid-b40 ratio. Prior to acceleration, however, there is already a pronounced synaptic dysfunction, reflected as changes in synaptic transmission and altered gene expression, indicating that restoring synaptic function early in the disease progression may represent the earliest possible target for intervention in the onset of Alzheimer’s disease.

The Arg-Gly-Asp (RGD) recognition site of platelet glycoprotein IIb-IIIa on nonactivated platelets is accessible to high-affinity macromolecules.
We have characterized a murine IgG monoclonal antibody, OP-G2, specific for platelet glycoprotein (GP) IIb-IIIa (alpha IIb beta 3). OP-G2 Fab fragments inhibit fibrinogen-mediated platelet aggregation and competitively inhibit adenosine diphosphate-induced binding of 125I-fibrinogen to washed platelets. OP-G2 binding to GPIIb-IIIa is specifically inhibited by RGD-containing peptides but not the...
متن کاملThe Traveler's Problem
The Traveler’s Problem (TP) entails determining the maximum distance that can be traversed along a road, given the locations and room rates of inns along that road, and given the constraints of maximum distance per day and limited budget available for overnight stays at inns. An OP(n 3 5_ (log n) 3 1_ ) time algorithm is presented for the TP. An OP(n 3 5_ (log n) 3 4_ )-time algorithm for compu...
متن کاملRobust Op Amp Realization of Chua's Circuit
Chua's circuit is a simple electronic network which exhibits a variety of bifurcation phenomena and attractors. The circuit consists of two capacitors, an inductor, a linear resistor, and a nonlinear resistor. This paper describes the design methodology for a robust practical op amp implementation of Chua's circuit. In addition, we present experimental results and SPICE simulations for a workin...

The effect of a chemically stable prostacyclin analog, OP-41483 alpha-cyclodextrin clathrate (OP-41483.alpha-CD), on vascular lesions, platelet aggregation and blood pressure were examined and compared with those of prostaglandin E1 alpha-cyclodextrin clathrate (PGE1.CD) in in vivo rat models. 1) In the laurate (1 mg/leg, i.a.)-induced arterial thrombotic model, OP-41483.alpha-CD (1 microgram/k...
متن کاملOsteopontin: a protective mediator of cardiac fibrosis?
Osteopontin (OP) is a multifunctional cytokine and adhesion protein that contains an RGD (argininglycin-aspartate) binding sequence that enables it to interact with several integrins, CD44 variants, and other adhesion receptors. OP receptor binding then directly or indirectly activates intracellular signaling pathways, mediating its effects on cell–matrix and cell–cell interactions. OP is incre...
متن کاملRat brain acetylcholinesterase response to monocrotophos and abate.
The exposure to organophosphorus (OP) pesticides lead to cholinesterase inhibition and cause an accumulation of acetylcholine in the central nervous system and other parts of the body (Gupta et al., 1971 & 1972). Inhibition of brain acetylcholinesterase (ACHE) is generally regarded as a useful indicator of poisoning by OP or carbamate pesticides (Martin et al., 1981; Grue et al., 1983). Depress...
